RSK1 is an exploitable dependency in myeloid malignancies Overall design: Here, we establish a comprehensive RNA-seq dataset of CD14+ cells encompassing 68 patient samples across the spectrum of myeloproliferative neoplasms with normal donor controls. CD14+ cells were bead-enriched from the CD34- sorted fraction from MPN PBMCs and normal healthy donor (bone marrow) to assess gene expression patterns.

Construction protocol: The CD34- cell fraction was then enriched for CD14+ cells with bead purification.Total RNA from cell lines was extracted with RNeasy Mini Kit (Qiagen, Hilden, Germany) with DNAse treatment following the manufacturer's protocol. Samples were then indexed, pooled, and sequenced by Illumina NovaSeq 6000 following standard Illumina protocol.
